Do you have to have been married for a certain amount of time to qualify for a Non Immigrant O Marriage visa, or can you do it literally as soon as you have the legal marriage certificate?

You can apply as soon as you are married.

Both for a Non Imm O Visa and a 12 month extension from Immigration providing meet the requirements.

Thanks LB. Also, another quick one, the first time you get a Non Immigrant O visa, do you have to leave the country and apply for it from a Thai Embassy or Consulate abroad, or can you do it all from within Thailand?

You can only obtain a non immigrant visa entry within Thailand if you qualify for one year extension of stay. If you do meet the requirements (400k in bank account or 40k per month income for marriage) and have sufficient time remaining on your current permitted to stay stamp you pay 2,000 baht for change (may have to be done in Bangkok) and later extend for one year for the normal 1,900 baht fee.
