I'll soon be leaving Thailand and changing from an O to a B visa, am I right in thinking I won't need a re-entry permit for my O visa as I'll be returning with a new B visa? But if there are issues with my non-imm B application I would still have 6 weeks of my non-imm O left so I could get a re-entry permit just in case there are any problems. I have a valid work permit which expires June 2016.

Yes you could get a re-entry permit (1,000 baht) before leaving Thailand which will keep the 6 week stay remaining alive.

Was the Non 'O' issued on the basis of marriage?
