I don't know for sure if you can get a mini van from Ubon Ratchathani to Yasothon but I can for 60 baht to Amnat Charoen and they leave every 15 minutes from the bus station.. Nakhon Chai Air bus service is far superior but goes to Ubon. I leave Pattaya (just south of Pattaya Klang on the Sukhumvit) and do the overnight trip and get a good sleep to boot. The drivers are very professional and safe plus they have a stewardess on board that delivers free snacks and a drink. The bus also has a washroom. It was about 650 baht for the deluxe bus. I hope that helps.
