very soon, on the 2nd of november. My 1st tourist visa entry expires, ( have been to cheng wattana soi 7 for 30 day extension already), but this is the first time that I have had a tourist visa with more than 1 entry granted- 3 entrys granted at queens gate Thai embassy, London UK.

So to use/activate the second entry, am I forced to go to Poi Pet, Cambodia, as I'm in Bangkok now, or can I just go to Suvannabhumi Airport or something instead.

Also if I do have to go to Poi pet will I have to get a Cambodia visa or can I just go to the Thai side.....

You cross any border and enter a neighbouring country, turn round and come back. Yes you will need a Visa if the country you enter requires one. For Cambodia I think it is $20 US.

You can go to the most convenient place for you, but you will have to exit Thailand and enter again.
