If my wife and I were in the U.K on holiday and I was offerd a job could we apply from the UK for her to stay?

At best you could get a few more months extension but I think it will cost a lot and I think it would be unlikely to succeed, so you could pay a lot in application fees and get nothing.

Your wife will need to return to Thailand to apply for "settlement" if she intends to live permanently in the UK with you, then she will be able to live, work and get all the other benefits that goes with being a permanent resident, can even apply for British nationality after 3 years.

Please remember as a visitor (VAF1A) she will not be able to work or be entitled to receive any form of benefits including NHS treatment, (though if she does require emergency treatment she will not be refused but will be expected to pay).

that was the short answer, there a few ways of getting a longer visitor visa like as student, etc but even then she would still need to return to Thailand eventually to apply for settlement.

The short answer is 'No.'

The long answer is that those in the UK with a non settlement visa valid for 6 months or less, e.g. a visit visa, cannot apply for settlement in the UK. She will have to return home to do so.
