Will be driving this route for the first time on Monday and will be alone so can anyone give me the quickest route for such a trip.

Bangkok head for Rangsit then pick up the A-2 heading for Ang-Thong stay on the A-2 passed Singburi onto Uthai Thani then all the way to Nakhon Sawan once there take the road Number 117 all the way to Phitsanulok

its about 380 klm

I made this drive yesterday... There are big signs at the Highway 32/Highway 11 junction in Singburi recommending using Highway 11 to both Phitsanulok and Phichit. I would not recommend taking Highway 11 unless you really want to drive on a two-lane road for 200+ km. We took Highway 11 just for ducks. Very little traffic but the road is fairly banged up in spots. Highway 117 is better (and four-lane).
