If my wife passed the former LIFE in UK (last year) to help get successful indefinite leave to remain visa for settlement, with the introduction of the new LIFE in UK test this year, will she need to take the new test as part of the application for British Passport?

She will have to get UK citizenship before she can get a UK passport.

If she is applying before 28th October 2013 then her LitUK test alone is sufficeint.

If she will be applying on or after 28th October 2013 then she will need a B1 or equivalent pass in English speaking and listening as well.
