using ghosting software, does the destination drive have to have the same capacity as the source drive or is a destination drive with enough capacity for the USED portion of the source drive sufficient?

Depends on the program's capabilities. Usually, if the capacity of the target drive/partition exceeds the source, then it's OK. Whether data exceeds and will work or not depends. If one of the drives is a seagate or Maxtor, then you can use the free software they provide on their website.
