Hello, what would happen if an immigration officer signs or stamps my ed visa sticker as used after I just got it in Laos? Will this cause a problem when I give my passport to the school and they try to get the 2nd approval letter from MOE?

If you have a single entry non-ed visa it should always be stamped used by immigration when you enter the country. This is normal procedure so you will not have a problem.

Your 90 day permit to stay you from the visa is what matters after you enter the country.

What if they forget to stamp used when Im at immigration?

Are you worrying about things that do not have even happened ?

Anyway, one should always check the stamps he/she gets. If any mistake is made, it will be corrected immediately.

It happens some times but not often. It would not matter for anything. You would still have your entry/permitted to stay stamp that shows you used your visa for entry.
