I need to drop into Poipet on my way from Isaan to Kho Samet, to get a 15 day visa extension.

Can I stamp out and straight back into Thailand, without having to actually go through Cambodia Immigration.

Nope.

You have to enter another country before coming back.

You must have entry/exit stamp from another country on a land border crossing.

Does any-one have info regards if a visa run can be made to Poipet during Songkran, especially Friday 16th. Is it open???

Borders are open every day, only immigration offices (where you go for extensions of stay etc). close for the holidays.
