First time visitor to Thailand and wanting to stay in Thailand for a year doing business, but I understand that Vientiane(where I am currently at) they will only issue a single entry 3 month Non-B visa. If I take this visa now will that prevent me from obtaining an extension for a total of one year's stay?

If not, will the Chiang Mai Immigration office be helpful in giving me all the information I will need to properly apply for the extension?

Also, what is the cost of the single entry non-B ?

Taking a single entry visa will have no effect on future visa issue. A Consulate does not extend your stay - only an immigration inside Thailand can do that and you need to meed work permit/income and other requirements. Working in Thailand requires a work permit - no visa permits work.
