Will enter Myanmar at Mandalay by flight and head down to Myawaddy.

3 pax: 2 UK and 1 Australian. The 2 UK people will want to enter at Myawaddy with a Thai Visa exemption issued at the Mae Sot Immigration point. The Aus has an O visa with multiple entry.

The question arose that UK passport holders can only depart Myanmar through an International airport. Is there any rule stating this? Is this true?

Not true at all. Since August 28, 2013 at 8am foreigners of all nationalities as well as Burmese have been allowed to enter/leave at any of 4 Thai - Myanmar checkpoints or airports at will.
