I would say the Nana for the girls and the Manhattan for cleanliness.

How far of a walk is it from Manhattan hotel to Nana plaza? What about from Manhattan hotel to Soi Cowboy? And from Manhattan to the beer garden? Is it a safe walk at 2:00am after having 6 beers?

The Manhattan sits about mid way between both points you mentioned. I'd say no more then 15 min., less if you're a fast walker. The beer garden (I assume you mean the Soi 7 beer garden) is between the Manhattan and the Nana so you can stop off there on your way to the Nana. I'd say you should be pretty safe but as usual take the normal precautions.
