I can play USB's but not Mini SD cards in a USB holder.

My car stereo has a USB port which works fine with a normal flash drive, but if I try and play a mini SD card that's inserted into a flash drive holder (so it can be used in a computer) it won't work. Why?

Because your stereo supports usb drives, but not your card reader, presumably.

I have found some card readers temperamental depending on the brand and capacity of the card. Did you try another, different brand card?

Only have Sandisk brand, so nothing else to try, but it's no biggie, I was just curious as to why. I can transfer all the music on the SD card to a flash drive easily enough.

Sandisk is my brand choice also. I had a card reader that normally had no problems reading them but bought a Ultra high speed Sandisk for my camera and it couldn't read it. Bought a new card reader and it could.
